The John Paul II Catholic University of Lublin
The competition is open to individuals who meet the requirements set out in Art. 113, 116 section 2 of the Act of July 20, 2018, Law on Higher Education and Science and meet the following qualification criteria:
The aim of the project is to discover the cellular and molecular basis of the antifungal activity of metallacarboranes. At the first stage of the research, the influence of metallacarboranes on the morphology and nanomechanical properties of fungal cells will be performed. Next, homeostasis (e.g. membrane permeability) and virulence factors (e.g. biofilm formation) of C. albicans cells treated with metallacarboranes will be assessed. Flow cytometry analysis is planned to study metallacarborane-mediated C. albicans cell death and the influence of the compounds on the cell cycle. The proposed study on the transcriptome of C. albicans using Next-Generation Sequencing followed by bioinformatic data analysis and Real-Time qPCR is aimed at assessment of changes in gene expression caused by metallacarboranes. Finally, the effect of the compounds on the proteome of the fungus will be studied, and an attempt will be undertaken to reveal the C. albicans protein interacting partner of the compounds using affinity chromatography.
